Slide Preparation: Wet Mount

Agar Culture

Place a loop full of water onto a clean glass slide. Up to three wet mounts can easily be placed on one slide, spacing them evenly.

Flame your loop, pick up a small amount of culture, and mix it into the drop of water without spreading it out too much.

Flame the loop and set it down. (See Aseptic Techniques: Flaming the Loop)

Place a clean cover slip over the liquid and press down lightly. If the liquid reaches the edge of the cover slip, you have used too much liquid and the cover slip will float off the slide under oil immersion.

Broth Culture

Mix the culture.
