Slide Preparation

There are many specific skills needed in the microbiological laboratory. These skills are used repeatedly in many different experiments or microbiological applications.

Because microbes are too small to be seen without magnification, we use a microscope to view them. In this section we will study how to prepare microbes to be viewed in the microscope, learning techniques to make slides (wet and dry) and to stain slides with a simple stain. Differential staining will be taught in your lab course. From visualizing the microbes you can tell their size and shape, whether they are prokaryotic or eukaryotic, how many kinds of organisms are in the sample, may provide an idea of how to treat an infection clinically, and the smear could provide clues as to how to identify the organism.
